I think Jacob and MIB are fighting for their birthright (Like the original bible bros). I think the island is a metaphor for mankind, and whoever's theories on mankind is correct, wins their birthright.
I think that they both need to be on the island to create balance, and so Jacob's insistence on protecting the island keeps the MIB trapped there. So Jacob is not really protecting the "island" but mankind, who he believes is good. MIB is done with mankind (they always come and fight) and is tired of playing this game and wants to go home... I think the candidates are the people that prove Jacob's theories on mankind and why they should be allowed to survive, and they are the only ones that can kill Jacob. Once all the candidates are gone, Jacob's theory is disproven and MIB can leave the island. Mankind is destroyed.
Also, the 4 toed statue looks like Egyptian God Anubis, (the jackal-headed god associated with mummification and the afterlife in Egyptian mythology). Anubis was the gate-keeper of the world of the dead. Anubis's job was to decide whether you lived forever or if you were eaten by a demon upon death. In order to decide this, after mummification he placed the deceased heart on a scale, if it was lighter than feather, you are golden... They also showed scales during this last episode, what's it mean? Besides balance and good vs evil.
